Description
We set up a white light off-axis digital holographic system using a 40× objective with a numerical aperture of 0.65. The central wavelength of the light souce is 574 nm. In this dataset, the sample consists of red blood cells from the glaucoma subjects and the healthy controls. The media is sodium chloride solution with the concentration of 0.7%. Digital holograms are recorded under different fields of view using a CMOS camera, with corresponding background holograms recorded for aberration correction. The exposure time for each hologram is 30000μs. For each subject, digital holograms of RBCs were acquired together with the corresponding background holograms recorded under identical optical conditions. The dataset enables quantitative phase imaging (QPI) studies, including phase reconstruction, background compensation, morphological analysis and the extraction of biophysical parameters such as cell thickness, volume and sphericity. It is designed to support research in biomedical optics, computational imaging and machine learning-based disease classification.
